چکیده

The helical stability of poly-L-glutamic acid and poly-Lornithine in aqueous alcohol solutions was studied by optical rotatory dispersion and potentiometric titration determinations. It was found that aliphatic alcohols enhance the stability of the helix according to the series methanol < ethanol < I-propanol < I-butanol. The role of secondary and tertiary alcohols was also investigated. Thermodynamic analysis of the thermal transition of uncharged poly-L-ornithine showed that the greater stability of the helix in aqueous alcohol solutions with respect to that in water arises from a more favorable value of the entropy change of the transition ASO. A similar effect of alcohols on the thermodynamics of the coil-helix transition in aqueous solution has already been observed for poly-L-glutamic acid. It is suggested that solvation of the peptide amide group in the randomly coiled conformation may be responsible for the observed trend of the variation of the thermodynamic quantities.

Volumetric characterization of homopolymeric amino acids.

We have determined the partial molar volumes, expansibilities, and adiabatic compressibilities for poly(L-alanine), poly(L-proline), and poly(L-threonine) within the temperature range of 18-55 degrees C. In addition, we have determined at 25 degrees C changes in volume, DeltaV, and adiabatic compressibility, DeltaK(S), associated with the coil-to-helix transitions of poly(L-lysine) and poly(L-g...

For several years, we have been engaged in evaluating the helix-coil stability constants for the naturally occurring amino acids in water.’ Specifically, we have determined the values of the parameters u and s of the Zimm-Bragg theory2 for several amino acids in aqueous solution, using the “host-guest” t e~hn ique .~ A similar series of studies to determine these same parameters in nonaqueous s...

Osmotic Pressure Induced Coupling between Cooperativity and Stability of a Helix-Coil Transition.

Most helix-coil transition theories can be characterized by three parameters: energetic, describing the (free) energy cost of forming a helical state in one repeating unit; entropic, accounting for the decrease of entropy due to formation of the helical state; and geometric, indicating how many repeating units are affected by the formation of one helical state. Depending on their effect on the ...

Crowding effect on helix-coil transition: beyond entropic stabilization.

We report circular dichroism measurements on the helix-coil transition of poly(L-glutamic acid) in solution with polyethylene glycol (PEG) as a crowding agent. The PEG solutions have been characterized by small angle neutron scattering and are well described by the picture of a network of mesh size ξ, usual for semi-dilute chains in good solvent. We show that the increase of PEG concentration s...
